Priciple of the procedure
The product adopts colloidal gold immunochromatography assay to qualitatively detect the novel coronavirus (SARS-CoV-2) neutralizing antibodies in human serum, plasma or whole blood samples.
In this product, colloidal gold labeled ACE2 is pretreated on the first layer of glass cellulose membrane, RBD antigen and chicken IgY are pre treated on the second layer of glass cellulose membrane, and ACE2 and goat anti-chicken IgY antibody are coated on the T and C lines of nitrocellulose membrane respectively.
During detection, if there is no neutralizing antibodies or its concentration is below the threshold value in the sample, the gold-labeled ACE2 can bind to the front pad RBD Ag, and when passing through the NC membrane, it can bind with the ACE2 on the membrane (o fomi a "ACE2-RBD Ag-ACE2" conjugate, thus a color band appears in the test area (T); And If the concentration of neutralizing antibodies in the sample is higher than the threshold value, the neutralizing antibodies will bind to RBD Ag, so as to competitively inhibit the binding of R.BD to ACE2 in the detection area, resulting in weaker bands or no bands in the test area (T) because of competitive inhibition.
Regardless of whether the measured substance is present in the sample, a band will appear in the control area (C). The band in the control area (C) can be used as control to compare with that of test area (T).
