5. Principles
Nitrofuran (AHD) rapid test strips are based on the principle of competitive inhibition immunochromatography. During the flow process, nitrofuran (AHD) in the sample binds to nitrofuran (AHD)-specific colloidal gold-labeled monoclonal antibody, inhibiting the antibody and nitrofuran (AHD)-BSA at the NC membrane detection line Associates coupling agent. pair combinations, resulting in a change in the color test line. When the sample has no nitrofuran (AHD) residue or the concentration is lower than the detection limit, the T line is darker than the C line or the T line is the same color as the C line; when the concentration is equal to or higher than the detection limit, the T line is obviously lighter than the C line or the T line Lines have no color. The C line appears regardless of whether nitrofuran (AHD) remains in the sample, indicating that the test is valid.
6. Sample Preparation
6.1 Solution Preparation
0.1MK2HPO4 solution: Dilute 1M K2HPO4 solution 1:9 with deionized water.
6.2 Sample Handling
Tissue:
1) Take 3±0.05g of homogenized tissue sample into a 50ml centrifuge tube, add 6ml of deionized water, 750ul of 1M HCl and 300ul of derivatization reagent, shake well for 2min, and take a 60°C water bath for 30min;
2) Add 7.5ml 0.1M K2HPO4 solution, 600ul 1M NaOH and 10ml ethyl acetate, shake well for 1min, and centrifuge at 4000r/min for 10min;
3) Take 5ml of supernatant in a glass tube and dry with nitrogen or air at 56°C.
4) Add 2ml of n-hexane, shake well to dissolve the dry residue (if it is not completely dissolved, the result may be unstable), then add 0.6ml of loading buffer, shake well for 30s, stabilize to separate layers, and then dry the upper layer (if there is an emulsion) ), centrifuged at 4000r/min for 5min);
5) Aspirate 200ul of the lower liquid (water layer) to prepare for testing. (Avoid absorbing upper liquid).
7. Operating procedures
7.1 Please read the manual carefully before use. Return the test strip and sample to room temperature.
7.2 Take out the required bottle from the kit, take out the required microwell and test paper, and make a mark. Please use these test strips within 1 hour. Seal lid to avoid moisture.
7.3 Take 200ul of the sample to be tested in the microwell, absorb it 5-10 times, mix the sample with the reagent in the microwell, until the naked eye judges that there is no solid (this is a very important step).
7.4 Incubate at room temperature (20-25°C) for 5 minutes, insert the test paper into the microwell, and completely immerse the "MAX" end in the mixed solution.
7.5 Incubate again at room temperature for 5-8min, read the result, other time is invalid.
8. Interpretation of test results
8.1 Negative: T line is darker than C line, or T line and C line are the same color. This means that there is no nitrofuran (AHD) in the sample or is below the detection limit.
8.2 Positive: T-line is significantly brighter than C-line or T-line is not visible. This means that the residual nitrofuran (AHD) is at or above the detection limit.
8.3 Unqualified: No wine red color can be seen on the C line. This means the test card is invalid, expired or mishandled. Please run the test again with another package. If invalid tests continue to occur, please contact the supplier.
9. Matters needing attention
1) The test card can only be used once at room temperature, please do not use the expired test card.
2) Do not reuse tips to avoid cross-contamination.
3) Do not touch the white film surface in the middle of the test card, avoid direct sunlight and direct fan blowing.
4) Wear gloves when testing.

10. Specificity
No cross-reactivity with nitrofurans AMOZ, AOZ and SEM.
