Testing Principle
The method is based on the pleochroic staining method of Lendrum (MSB method), and the staining is
completed by using an anionic dye mixed or sequentially. Different tissue components can be displayed by
selecting different anionic dyes of different molecular sizes for staining according to different permeation
properties of the tissue. That is, the densely-packed red blood cells are selectively stained with a small molecular weight of Matthew, and then the cellulose and muscle fibers are stained red with a medium molecular weight acid fuchsin. Finally, the loosely-structured collagen fibers are stained blue with a large molecular weight aniline blue.
