Testing Principle
The kit methodology is based on the standard Ziehl-Neelsen method recommended by the World Health Organization (WHO) and is mainly used for chemical staining of acid-fast bacteria such as Mycobacterium tuberculosis.
Acid-resistant bacteria such as tuberculosis and bacillus are not easily colored because of a layer of lipid or lipid on the surface of the cells, but once colored, the effect of acidic alcohol is not easy to decolorize. Using this property and staining with an enhanced staining solution, and then treating it with acidic alcohol, decolorizing it and then comparing the staining. At this time, the acid-resistant bacteria are still fixed with the originally stained color (red), which is easy to identify.
Stain Results
In light blue background, the acid-fast bacteria are red, and other bacteria and cells are blue.
Most Mycobacterium tuberculosis is bacilli and slightly curved. The width of bacteria (0.3~0.6) μm, and the length of bacteria is different, between (0.5~8) μm, most of them are (1.5~3.5) μm. A small number of Mycobacterium tuberculosis may be spiral, and the darker coloring metachromatic granules can be found in the well stained Mycobacterium tuberculosis.
