Testing Principle
This product is formulated based on a novel staining technology researched by Professor Liu Zhenhui from National Taiwan University. Similar to Wright's stain, it is an improvement derived from the Romanowsky stain technique, yielding results akin to those of Wright's stain. However, Liu's staining method requires a shorter duration, typically completing the staining process in just 1 to 2 minutes.
The cell staining process involves the penetration and retention of dyes within the stained material, a process that encompasses both physical adsorption and chemical affinity. Due to the differing chemical properties of various cells and their components, their affinity towards the acidic dye (eosin) and basic dye (methylene blue) in Liu's staining solution varies. Consequently, after staining with Liu's staining solution, different types of cells exhibit distinct colorations, thereby facilitating the identification of their morphology and characteristics.
Stain Results
1 Blood Smear and Bone Marrow Smear Staining:
Red blood cells appear light red or light green.Granules in the cytoplasm of white blood cells are clearly visible, displaying the unique colors characteristic of each cell type.Cell nuclei are stained purple-red, with clearly visible chromatin structures.
2 Gynecological Discharge Staining:
①Trichomonads: After staining, trichomonads mostly appear pear-shaped, round, oval, or irregular. The cytoplasm is stained gray-blue or sky-blue, with a small, date pit-shaped nucleus often in one side. Careful observation is required to avoid confusion with epithelial cells. Flagella may occasionally be visible.
②Candida (Yeast): Stained dark blue, with visible budding spores and slender, straight pseudohyphae.
③White Blood Cells: Morphologically identical to white blood cells in blood smears. The quantity of white blood cells generally indicates the degree of inflammation, providing a useful indicator for physician diagnosis.
④Leptothrix: Long and thin, appearing gray-blue.
⑤Gonococci: Blue, typically kidney-shaped and often found inside or outside white blood cells and tissue cells. Note that staphylococci commonly appear in female vaginal smears, with staining results similar to gonococci, also appearing in pairs as diplococci. Therefore, interpretation of gonococci requires great caution.⑥Gardnerella: Blue, coccobacillary in shape.
3 Exfoliated Cell Staining:
Cell nuclei are stained purple-red, and nucleoli are stained blue.
The cytoplasm exhibits varying shades of blue depending on the degree of differentiation, reflecting changes in basophilia.
