Diff-Quik Stain (For Auto Stainer) Baso Clear Colourful Stains
Testing Principle
Proteins within cells and sperm, which have different isoelectric points, carry varying charges under the same acidity conditions, enabling them to selectively bind to corresponding dyes and become stained. Acidophilic proteins dissociate to form amino groups with positive charges, which can bind to the negatively charged acidic dye (eosin) and appear red. Basophilic proteins dissociate to form carboxyl groups with negative charges, which can bind to the positively charged basic dye (methylene blue) and appear blue. Neutrophilic proteins dissociate to form equal amounts of positively charged amino groups and negatively charged carboxyl groups, simultaneously binding equal amounts of acidic and basic dyes, resulting in a purple-red color. However, due to the equal dissociation of charges, the staining is relatively weak. This staining method is the WHO-recommended DiOuik staining technique.
Stain Results
1.Blood Smear Staining
Red blood cells appear red,Granules in the cytoplasm of white blood cells are clearly visible, displaying the unique colors characteristic of each cell type. Cell nuclei are stained purple-red, with clearly visible chromatin structures.
2.Semen Smear Staining
The post-acrosomal region of the sperm head appears purple-blue, while the acrosomal region appears light purple-red.The midpiece and tail of the sperm are stained light red.Various cells in the semen exhibit different morphologies and staining results due to their distinct shapes and compositions.
