Testing Principle
Sperm and proteins within cells, which have different isoelectric points, carry varying charges under the same acidity conditions, enabling them to selectively bind to corresponding dyes and become stained. The nuclei of sperm and cells are primarily composed of acidic substances, making them stainable with the basic dye hematoxylin. Conversely, the cytoplasm mainly contains basic substances, which have a greater affinity for acidic dyes. This staining method is a simplified version of the Papanicolaou (Pap) stain, modified based on the operational characteristics of the accompanying staining machine, and builds upon the WHO-recommended standard Pap staining technique.
Stain Results
The acrosomal region of the sperm head is stained light blue or appears colorless and transparent, while the post-acrosomal region is stained dark blue. The midpiece and tail of the sperm are stained light red or light green. Various cells in the semen exhibit different morphologies and staining results due to their distinct shapes and compositions.
